Reduced pyrazinamidase activity and the natural resistance of Mycobacterium kansasii to the antituberculosis drug pyrazinamide.

Pyrazinamide (PZA), an analog of nicotinamide, is a prodrug that requires conversion to the bactericidal compound pyrazinoic acid (POA) by the bacterial pyrazinamidase (PZase) activity of nicotinamidase to show activity against Mycobacterium tuberculosis. Mutations leading to a loss of PZase activity cause PZA resistance in M. tuberculosis. M. kansasii is naturally resistant to PZA and has redu...

Crystal Structure of the Pyrazinamidase of Mycobacterium tuberculosis: Insights into Natural and Acquired Resistance to Pyrazinamide

Pyrazinamidase (PncA) activates the first-line antituberculous drug pyrazinamide into pyrazinoic acid. The crystal structure of the Mycobacterium tuberculosis PncA protein has been determined, showing significant differences in the substrate binding cavity when compared to the pyrazinamidases from Pyrococcus horikoshii and Acinetobacter baumanii. In M. tuberculosis, this region was found to hol...

Susceptibility of Mycobacterium tuberculosis to pyrazinamide and its relationship to pyrazinamidase activity.

Pyrazinamidase activity has been associated with pyrazinamide-susceptible Mycobacterium tuberculosis strains. The detection of pyrazinamidase activity by the Wayne method was found to be of limited value when compared with the results of standard pyrazinamide susceptibility tests, especially when a high level of pyrazinamide resistance was found. When resistance to pyrazinamide reached a level ...

Assessment of adherence to antituberculosis drug regimen among patients receiving isoniazid, rifampin and pyrazinamide: Kerman, Iran

Introdution: Among infectious diseases, tuberculosis is one of the leading causes of death killing in nearly 1.5 million people yearly. Considering the importance of patient co-operation in drug therapy and resistance we decided to evaluate the rate of co-operation of tuberculosis infected patients in regard to their use of anti-tuberculosis drugs in Kerman. Methods: This descriptive study was...

Primary drug resistance of Mycobacterium tuberculosis to antituberculosis drugs.

Thirty two positive sputa from 104 untreated cases of pulmonary tuberculosis, were tested for sensitivities against antituberculosis drugs. Twenty four were positive on direct smear as well as culture while 8 were only culture positive. Nine (28%) were resistant to Isoniazid, 3 (9.4%) to Streptomycin and 2 (6.3%) were resistant to both the drugs. All the 32 were sensitive to Rifampicin and Etha...
